Is it possible to alter an image using the Chroma-Key affect more then once, and if so, how is it done?

Not in a single pass. What you can do is use the chroma-key effect as desired and then produce to a file. In step 2, import the produced file into PD and use the chroma-key effect again as desired.

That's the current method I've been using; but I thought the ability to do it in a single-pass was available. Thanks

I assume it's a still image.

My advice would be to edit the image separately before bringing it into PD... which is not a graphics editor.

I use Paint.net to put transparency into images. I find it gives finer control of tolerance levels - and therefore better control of the look of the final image in the PIP track. When the image is ready, save it as a png file and it will retain whatever transparency you assigned it.
